[Abstract]Objective To targeted collect the biological samples of patients with cerebral apoplexy in Qiqihar,to construct a standardized biological sample bank of cerebral apoplexy with high quality and provide sample sources and data supporting platform for the data analysis and clinical decisions for cerebral apoplexy in The standardized operative process was made,the object was selected,blood serum,blood plasma,white cells,DNA and RNA extracted from the white cells of the standardized patients were collected and stored in the refrigerator at-80℃or liquid quality of DNA and RNA was detected so that clinical sample sources with high quality were to be From 2013,a total of 753 patients were enrolled in the sample of blood serum and blood plasma were 3008 respectively,white cells were 2253 and DNA and RNA were 3770 results of electrophoresis showed the RNA electrophoresis had both 18S and 28S stripes without dispersion and the lumen of 28S stripe was as twice as that of the 18S electrophoresis had clear stripes with good The standardized biological sample banks of cerebral apoplexy with high quality in Qiqihar is primarily established to further explore and apply the data sources of cerebral apoplexy,which can provide help for individualized treatment of patients with cerebral apoplexy in the future.

脑卒中是齐齐哈尔地区的高发疾病,严重危害人类健康,具有高发病率、高复发率、高致残率和高致死率的特点[1]。2008年原卫生部发布的《我国居民第三次死因回顾抽样调查报告》指出,脑血管疾病已成为导致我国居民死亡的首要原因,其中以脑卒中造成的损害为重[2],给患者家庭和社会带来了沉重的负担。
近年来,临床研究人员逐渐意识到样本资源对于科研工作的重要性,开始重视临床生物样本的大量收集和规范化的管理工作[3-5]。从2013年起,本院开始收集样本,建立脑卒中患者的生物样本库,旨在为今后的医学研究提供珍贵的临床样本资源。样本库的建立有助于探索脑卒中的发病机制,为脑卒中的临床治疗提供帮助。
1 材料与方法
1.1主要设备
小液氮罐,-80℃超低温冰箱(三洋MDF-U73V型),低温高速离心机(Thermo MICRO21R型),纯水机(Milli-Q Reference),生物安全柜(海尔HR60-iiA2型),凝胶成像系统(UVP GelDoc-ITTS2型),微量核酸蛋白检测仪(ND2000型),涡旋振荡器(Vortex-Genie 2T型)。
1.2主要试剂
PBS缓冲液(北京索莱宝科技有限公司P1010),红细胞裂解液(QIAGEN),DNA提取试剂盒(碧云天生物技术研究所D0063),Trizol(Thermo Fisher -026),样本稳定剂(Ambion AM7020),氯仿(上海金穗生物科技有限公司67-66-3),异丙醇(济宁铭达新材料有限公司67-63-0),80%乙醇(济宁华凯树脂有限公司64-17-5)。
1.3研究对象
以填表的形式对脑卒中患者进行流行病学调查,调查内容如下。①患者的一般情况:姓名、年龄、性别、民族、居住地址、文化程度及工作类别等;②疾病史:高血压、冠心病、肾病、糖尿病、脑卒中等既往病史及家族病史;③生活习惯:是否吸烟、喝酒或长期食用高盐或腌制品等;④临床信息:患者主诉、脑卒中亚型、并发症以及各类卒中相关评分等,根据调查情况确定入库对象。
1.4血液样本的采集及处理
筛选符合标准的脑卒中患者,签署知情同意书,采集患者空腹静脉血至抗凝管及非抗凝管中,2000 r/min离心10 min,吸取上清液分装,上清液分别为血浆和血清,置于-80℃冰箱或液氮中储存。取抗凝管中层液体到10 ml离心管中,加入5 ml红细胞裂解液,置于摇床充分混匀,2000 r/min离心10 min,弃去上清,重复2次,所得沉淀用PBS洗涤并分装,即得白细胞,加入样本稳定剂,4℃过夜充分渗透后冻存于-80℃冰箱或液氮中。
1.5 RNA和DNA的提取及鉴定
1.5.1 RNA提取及鉴定 1 ml Trizol吹散白细胞沉淀,加入200 μl氯仿离心20 min,吸取上清到装有560 μl冷异丙醇的EP管中混匀,常温静置1 h,4℃,12 000×g离心10 min,弃去废液,加入1 ml 80%乙醇,8000 g离心3 min,弃去废液,重复该步骤一次,所得沉淀即为RNA,用RNase-free H2O溶解,分装后置于-80℃冰箱或液氮中冻存。
取1 μl提取后产物,用微量核酸蛋白检测仪检测RNA的浓度及纯度,1%琼脂糖凝胶电泳后观察其完整性。
1.5.2 DNA提取及鉴定 DNA提取方法按DNA提取试剂盒说明书进行,产物分装后置于-80℃冰箱或液氮中冻存。
取1 μl提取后产物,用微量核酸蛋白检测仪检测DNA的浓度及纯度,2%琼脂糖凝胶电泳后观察其完整性。
